Biodeterioration patterns found in dammar resin used as art material Electron. J. Biotechnol.
Romero-Noguera,Julio; Martín-Sánchez,Inés; López-Miras,María del Mar; Ramos-López,José Miguel; Bolívar-Galiano,Fernando.
Since the middle of the XIX century, when dammar became popular in Occident, this natural resin is one of the most used in art painting techniques as final protective coating (varnish) as well as a component of pictorial media. The present work is the first approach to the study of the microbiological biodeterioration of this artistic material, which can seriously affect the appearance and integrity of works of art when bad conservation conditions -especially high humidity levels- take place. 12 microorganisms, fungi and bacteria, came from collection and from oil paintings affected by biodeterioration patterns, were inoculated on test specimens prepared with varnish dammar. These were incubated and analyzed by GC-MS to determine both the microbiological...

A microbial community analysis of the octocoral Eunicea fusca Electron. J. Biotechnol.
Duque-Alarcón,Angela; Santiago-Vázquez,Lory Z; Kerr,Russell G.
While there is a significant and growing body of knowledge describing the microbial communities of marine invertebrates such as sponges, there are very few such studies focused on octocorals. The octocoral Eunicea fusca is common on reefs in various regions of the Caribbean and has been the subject of natural product investigations. As part of an effort to describe the microbial community associated with octocorals, a culture-independent analysis of the bacterial community of E. fusca was conducted. Specifically, a 16S rDNA clone library analysis was performed to provide baseline data. A total of 40 bacteria members from 11 groups were found. In general, Proteobacteria were the dominant group with a total of 24 species and α-Proteobacteria...

Comparison of commercially-available RNA extraction methods for effective bacterial RNA isolation from milk spiked samples Electron. J. Biotechnol.
García-Nogales,Paula; Serrano,Alicia; Secchi,Sonia; Gutiérrez,Serafín; Arís,Anna.
Nucleic-acid based methods for bacterial identification are extremely useful in diagnostic applications due to their specificity and sensitivity. However, they require an optimal purification of the target molecules. As part of the development of a new diagnostic method for the detection of bacterial RNA in cow milk, we have compared four commercially available RNA extraction kits for the isolation of bacterial RNA from spiked UHT milk samples. The kits were compared in terms of extraction efficiency and RNA purity using two bacterial species, the Gram negative Escherichia coli and the Gram positive Staphylococcus aureus. Two kits are based in silica-matrix extraction, and the other two in the guanidinium thiocyanate-phenol-chloroform extraction. In our...

Silanol - A novel class of antimicrobial agent Electron. J. Biotechnol.
Kim,Yun-mi; Farrah,Samuel; Baney,Ronald H..
Recently, a significant amount of attention has been directed toward development of novel classes of biocides because of the potential for microbial contamination and infection risks to military personnel and in the general population. We have recently discovered a new and unexpected class of powerful biocides based upon compounds derived through simple chemistry from silicone intermediates, "silicon alcohols" called, silanols. One example is trialkylsilanols, R3SiOH. Antimicrobial tests were carried out against Gram-negative bacterium, Escherichia coli, and Gram-positive, Staphylococcus aureus, with silanols, t-butanol, and siloxanes. Trialkylsilanols were very effective biocides against Escherichia coli and Staphylococcus aureus. The number of viable...

Novel antimicrobial activity of a dichloromethane extract obtained from red seaweed Ceramium rubrum (Hudson) (Rhodophyta: Florideophyceae) against Yersinia ruckeri and Saprolegnia parasitica, agents that cause diseases in salmonids Electron. J. Biotechnol.
Cortés,Yurima; Hormazábal,Emilio; Leal,Hellmuth; Urzúa,Alejandro; Mutis,Ana; Parra,Leonardo; Quiroz,Andrés.
Background Enteric red mouth disease and Saprolegniasis, which are caused by the bacteria Yersinia ruckeri and the oomycete Saprolegnia parasitica, respectively, are important illnesses that affect salmonid farming. Sanitary problems in farms are addressed by the prevention of disease outbreaks or by the treatment of diseases with chemicals. Environmental and governmental restrictions, toxicity and high treatment costs limit the use of drugs. Marine organisms, such as algae, sponges and corals, have developed an antimicrobial defense strategy based on the production of bioactive metabolites. Among these organisms, seaweeds offer a particularly rich source of potential new drugs. Hence, many pharmacologically active substances have been isolated from...

Leaf litter decomposition from native and non-native species in a freshwater forested wetland of Chile Gayana
Gómez-Capponi,Francisco; Correa-Araneda,Francisco; Díaz,Ma Elisa; Olguín,Manuel; Encina-Montoya,Francisco; Figueroa,Ricardo.
ABSTRACT Decomposition of leaf litter is a fundamental process for the functioning of forested wetlands. The rapid increment of forest plantations has involved a greater contribution of leaf litter of exotic origin to these ecosystems. The decomposition rate between leaf litter of native and exotic origin in a forested wetland of the Mediterranean zone of Chile was compared, as well as the contribution of bacteria and macroinvertebrate to this process. It was determined that the decomposition rate of the leaf litter presented significant differences (p < 0.05) between species, being faster in those of non-native origin. This could be due to differences in ecophysiological aspects of the leaves like the presence of chemical compounds such as waxes or...

Leaf-associated bacterial microbiota of coffee and its correlation with manganese and calcium levels on leaves Genet. Mol. Biol.
Sousa,Leandro Pio de; Silva,Marcio José da; Costa Mondego,Jorge Maurício.
Abstract Coffee is one of the most valuable agricultural commodities and the plants’ leaves are the primary site of infection for most coffee diseases, such as the devastating coffee leaf rust. Therefore, the use of bacterial microbiota that inhabits coffee leaves to fight infections could be an alternative agricultural method to protect against coffee diseases. Here, we report the leaf-associated bacteria in three coffee genotypes over the course of a year, with the aim to determine the diversity of bacterial microbiota. The results indicate a prevalence of Enterobacteriales in Coffea canephora, Pseudomonadales in C. arabica ‘Obatã’, and an intriguing lack of bacterial dominance in C. arabica ‘Catuaí’. Using PERMANOVA analyses, we assessed the association...

Characterization of a small cryptic plasmid from endophytic Pantoea agglomerans and its use in the construction of an expression vector Genet. Mol. Biol.
Rudi Emerson de Lima,Procópio; Araújo,Welington Luiz; Andreote,Fernando Dini; Azevedo,João Lúcio.
A circular cryptic plasmid named pPAGA (2,734 bp) was isolated from Pantoea agglomerans strain EGE6 (an endophytic bacterial isolate from eucalyptus). Sequence analysis revealed that the plasmid has a G+C content of 51% and contains four potential ORFs, 238(A), 250(B), 131(C), and 129(D) amino acids in length without homology to known proteins. The shuttle vector pLGM1 was constructed by combining the pPAGA plasmid with pGFPmut3.0 (which harbors a gene encoding green fluorescent protein, GFP), and the resulting construct was used to over-express GFP in E. coli and P. agglomerans cells. GFP production was used to monitor the colonization of strain EGE6gfp in various plant tissues by fluorescence microscopy. Analysis of EGE6gfp colonization showed that 14...

Wolbachia in guilds of Anastrepha fruit flies (Tephritidae) and parasitoid wasps (Braconidae) Genet. Mol. Biol.
Mascarenhas,Rodrigo O; Prezotto,Leandro F; Perondini,André Luiz P; Marino,Celso Luiz; Selivon,Denise.
Abstract The endosymbiont Wolbachia is efficiently transmitted from females to their progenies, but horizontal transmission between different taxa is also known to occur. Aiming to determine if horizontal transmission might have occurred between Anastrepha fruit flies and associated braconid wasps, infection by Wolbachia was screened by amplification of a fragment of the wsp gene. Eight species of the genus Anastrepha were analyzed, from which six species of associated parasitoid wasps were recovered. The endosymbiont was found in seven Anastrepha species and in five species of braconids. The WSP Typing methodology detected eight wsp alleles belonging to Wolbachia supergroup A. Three were already known and five were new ones, among which four were found to...
