Since the middle of the XIX century, when dammar became popular in Occident, this natural resin is one of the most used in art painting techniques as final protective coating (varnish) as well as a component of pictorial media. The present work is the first approach to the study of the microbiological biodeterioration of this artistic material, which can seriously affect the appearance and integrity of works of art when bad conservation conditions -especially high humidity levels- take place. 12 microorganisms, fungi and bacteria, came from collection and from oil paintings affected by biodeterioration patterns, were inoculated on test specimens prepared with varnish dammar. These were incubated and analyzed by GC-MS to determine both the microbiological... |

Background: In the past decade, considerable attention has been paid for the development of novel strategies for the synthesis of different kind of nano-objects. Most of the current strategies are usually working by the use physical or chemical principles to develop a myriad of nano-objects with multiple applications. Main fields of nanotechnology applications range from catalysis, micro- and nano-electronics (semiconductors, single electrons transistors), non-linear optic devices, photo-electrochemistry to biomedicine, diagnostics, foods and environment, chemical analysis and others. Results: Two main avenues for nanoparticles synthesis: cell-free extract and cell cultivation have been reported. The state of art of both biotechnological approaches for... |

Nucleic-acid based methods for bacterial identification are extremely useful in diagnostic applications due to their specificity and sensitivity. However, they require an optimal purification of the target molecules. As part of the development of a new diagnostic method for the detection of bacterial RNA in cow milk, we have compared four commercially available RNA extraction kits for the isolation of bacterial RNA from spiked UHT milk samples. The kits were compared in terms of extraction efficiency and RNA purity using two bacterial species, the Gram negative Escherichia coli and the Gram positive Staphylococcus aureus. Two kits are based in silica-matrix extraction, and the other two in the guanidinium thiocyanate-phenol-chloroform extraction. In our... |

Recently, a significant amount of attention has been directed toward development of novel classes of biocides because of the potential for microbial contamination and infection risks to military personnel and in the general population. We have recently discovered a new and unexpected class of powerful biocides based upon compounds derived through simple chemistry from silicone intermediates, "silicon alcohols" called, silanols. One example is trialkylsilanols, R3SiOH. Antimicrobial tests were carried out against Gram-negative bacterium, Escherichia coli, and Gram-positive, Staphylococcus aureus, with silanols, t-butanol, and siloxanes. Trialkylsilanols were very effective biocides against Escherichia coli and Staphylococcus aureus. The number of viable... |

Background: Cellulases and lipases have broad industrial application, which calls for an urgent exploration of microorganisms from extreme environments as valuable source of commercial enzyme. In this context, the present work describes the bioprospection and identification of deep-sea bacteria that produce cellulases and lipases, as well their optimal temperature of activity. Results: The first step of this study was the screening of cellulolytic and lipolytic deep-sea bacteria from sediment and water column, which was conducted with substrates linked with 4-Methylumbelliferyl. Among the 161 strains evaluated, 40 were cellulolytic, 23 were lipolytic and 5 exhibited both activities. Cellulolytic and lipolytic bacteria are more common in sediment than at... |

Background Enteric red mouth disease and Saprolegniasis, which are caused by the bacteria Yersinia ruckeri and the oomycete Saprolegnia parasitica, respectively, are important illnesses that affect salmonid farming. Sanitary problems in farms are addressed by the prevention of disease outbreaks or by the treatment of diseases with chemicals. Environmental and governmental restrictions, toxicity and high treatment costs limit the use of drugs. Marine organisms, such as algae, sponges and corals, have developed an antimicrobial defense strategy based on the production of bioactive metabolites. Among these organisms, seaweeds offer a particularly rich source of potential new drugs. Hence, many pharmacologically active substances have been isolated from... |

The strain Chryseobacterium sp. kr6 shown to be useful for biotechnological purposes such as hydrolysis of poultry feathers and de-hairing of bovine pelts. The effect of temperature, initial pH and media composition on protease production by this keratinolytic strain was studied. The enzyme was produced between 25 and 37ºC, with maximum activity and yield at 30ºC. When protease production was tested in media with different initial pH, maximum activity was observed when cultivation was carried out at 30ºC and initial pH ranging from 6.0 to 8.0. Higher activity was observed when feathers or feather meal were used as growth substrates, followed by soybean meal. The addition of carbohydrates or surfactants to feather broth resulted in decrease in keratinolytic... |

Abstract Coffee is one of the most valuable agricultural commodities and the plants’ leaves are the primary site of infection for most coffee diseases, such as the devastating coffee leaf rust. Therefore, the use of bacterial microbiota that inhabits coffee leaves to fight infections could be an alternative agricultural method to protect against coffee diseases. Here, we report the leaf-associated bacteria in three coffee genotypes over the course of a year, with the aim to determine the diversity of bacterial microbiota. The results indicate a prevalence of Enterobacteriales in Coffea canephora, Pseudomonadales in C. arabica ‘Obatã’, and an intriguing lack of bacterial dominance in C. arabica ‘Catuaí’. Using PERMANOVA analyses, we assessed the association... |

A circular cryptic plasmid named pPAGA (2,734 bp) was isolated from Pantoea agglomerans strain EGE6 (an endophytic bacterial isolate from eucalyptus). Sequence analysis revealed that the plasmid has a G+C content of 51% and contains four potential ORFs, 238(A), 250(B), 131(C), and 129(D) amino acids in length without homology to known proteins. The shuttle vector pLGM1 was constructed by combining the pPAGA plasmid with pGFPmut3.0 (which harbors a gene encoding green fluorescent protein, GFP), and the resulting construct was used to over-express GFP in E. coli and P. agglomerans cells. GFP production was used to monitor the colonization of strain EGE6gfp in various plant tissues by fluorescence microscopy. Analysis of EGE6gfp colonization showed that 14... |
